Caffeine (CAS #58-08-2, > 99% purity) was purchased from SigmaAldrich Co., Ltd. (St. Louis, MO, USA). Isoflurane was purchased from Baxter Healthcare Co. (Deerfield, IL, USA). Safranin O (CAS No. 477-73-6) and Fast Green (CAS No. 2353-45-9) were obtained from Hengyuan Biotech (Shanghai, China). A TRIzol reagent (SKU: R6830-02) was purchased from Omega Bio-Tek (Doraville, GA, USA). Reverse transcription and quantitative PCR kits were purchased from Takara Biotechnology Co., Ltd. (Dalian, China). Oligonucleotide primers for the gene used for analysis were synthesized by Sangon Biotech Co., Ltd.(Shanghai, China). RIPA lysis buffer and a BCA Protein Assay kit were purchased from Beyotime Institute of Biotechnology (Haimen, China).Western Bright ECL HRP substrate was purchased from Advansta (Menlo Park, CA, USA). Fetal bovine serums were supplied by Gibco (St.Louis, MO, USA). Recombinant Murine IGF1 (250-19) was purchased from PeproTech, Inc. (Rocky Hill, NJ, USA). Advanced glycation end products (AGE-BSA, 2221-BSA) and BSA (2221-10) were purchased from BioVision (Milpitas, CA, USA). Low molecular weight heparin (LMWH) was purchased from Sigma-Aldrich Co., Ltd. (St. Louis, MO,USA). Collagenase II was obtained from Invitrogen (Carlsbad, CA, USA).Polyclonal antibody information is as follows: glyceraldehyde 3-phosphatedehydrogenase (GAPDH) (No. AC033), anti-β-actin (No.AB_2768234), α1 chain of type II collagen gene (COL2A1) (No.ABP51023), aggrecan (ACAN) (No. bs-1223R), GLUT1 (No. 21829-1-AP), AGEs (No. bs-1158R), ADAM metallopeptidase with thrombospondin type 1 motif 5 (ADAMTS-5) (No. ab41037), matrix metallopeptidase 13 (MMP13) (No. ab39012), IGF1 (No. sc-9013), and acetylhistone 3 lysine 9 (H3K9) (No. A7255). Secondary antibody information is as follows: Cy3 conjugated goat anti-rabbit IgG (H + L) (No.B100801) and Cy3 conjugated goat anti-rabbit IgG (H + L) (No. AS-1111). The other chemicals and agents were analytical grade.
